“It Didn’t Work on Me” #why. Source- https://doi.org/10.1186/s13073-026-01630-0 A recent study published in Genome Medicine reveals that genetic variants in the PAM gene, present in roughly ten percent of the population, can significantly reduce a person's response to GLP-1 medications. The PAM gene produces an enzyme responsible for activating metabolic hormones. Researchers discovered that individuals with these specific variations actually have higher circulating levels of the hormone but exhibit a strong biological resistance to it. As a result, the hormone and medications designed to mimic it become far less effective at slowing digestion and regulating blood sugar. When evaluating clinical trial data, the researchers found that patients carrying these PAM variants experienced up to a 44 percent reduction in the therapeutic benefits of GLP-1 receptor agonists compared to those without the variant. This unexpected resistance explains why a significant portion of individuals with type 2 diabetes do not see the anticipated improvements from these widely prescribed treatments. #health #glp1 #weightloss
